"How far can one use n.m.r. to determine the complete three-dimensional structure, of an enzyme in solution ? Can n.m.r. studies help to identify functional groups involved in catalysis, the nature of the transition state, and the role of any metal ions and co factors, etc ? What are the limitations of n.m.r. and in cases wh ere structural information of the enzyme itself cannot be obtained, can n.m.r. help ? Can the stereochemical relationships between ligand sites on allosteric enzymes be determine ? What aspects of n.m.r. are sensitive to different conformational states of an enzyme ? How do the use of paramagnetic probes and chemical modifications make the solution of these problems easier ?
This book attempts to answer some of these questions-and by suitable examples tries to discuss and assess the information that the different types of magnetic resonance measurements can give.
The use of paragmagnetic ions as probes features prominently in this book, and the section on this is intended to serve as a handbook to the biochemist and biophysicist. A brief introduction on the use of spin labels is also included. Because the methods described are applicable to most systems this book will also appeal to the chemist-particularly those whose studies involve the use of paramagnetic ions." |
